Kofu is about 4 hours from Osaka (https://goo.gl/maps/t5SDVh4MmahXC7Jk7), so it might be better to stay overnight… but that’s up to you 🙂
As a quick check, the local bus company says on their website that while they are moving towards accessible buses, step-only buses are still in operation and suggest calling the bus company the day before if you want to check a certain route.
